Restriction enzymes are used to recognize a specific sequence of nucleotides and cut double stranded DNA into smaller fragments at a specific location. Each restriction enzyme recognises different DNA sequences. HpaI is a Restriction Endonuclease purified from an Escherichia coli strain that carries the HpaI gene from Haemophilus parainfluenzae. HpaI activity is blocked by some combinations of overlapping CpG methylation. The optimum reaction temperature is at 37 °C.

Concentration: 10000 U/mL

Reaction Buffer: NZYtech provides HpaI with both a specific buffer, NZYBuffer B. For double digestions please use 10x NZYBuffer U (MB110, not provided).

Heat inactivation: No. The DNA should be purified with a phenol extraction or with a suitable DNA purification kit, such as NZYGelpure (MB011), if the enzyme is to be removed.
